Amazonian açai and food dyes for staining arbuscular- micorrhizal fungi PFB - Pesquisa Florestal Brasileira
Silva, Aline Lourdes Martins; Santana, Marcos Diones Ferreira; Pereira, John César de Jesus; Raimam, Milena Pupo; Albino, Ulisses Brigatto.
Arbuscular mycorrhizae microscopy requires differential staining of typical structures. Dyes employed, such as trypan blue, pose risks to health and environment. Alternative dyes such as pen ink and aniline have variable coloring efficiency. In this work, Brachiaria decumbens roots, discolored with caustic soda (NaOH), were stained with açai, annatto, saffron, trypan blue and pen inks. There were significant differences among dyes regarding stained mycorrhizal structures and pictures quality. Acai was considered the best alternative dye, with similar results to trypan blue.

Arbuscular mycorrhizal fungal communities in soils under three phytophysiognomies of the Brazilian Atlantic Forest Acta Botanica
Duarte,Lorrane Marques; Bertini,Simone Cristina Braga; Stürmer,Sidney Luiz; Lambais,Marcio Rodrigues; Azevedo,Lucas Carvalho Basilio.
ABSTRACT Arbuscular mycorrhizal fungi (AMF) play an important role in plant community productivity and structure, and so studying the factors that affect the diversity and structure of this fungal community is important for understanding their ecology in tropical forests. We investigated AMF spore communities and root colonization under three forest phytophysiognomies (Restinga Forest, REF; Lowland Ombrophilous Dense Forest, LLF; and Montane Ombrophilous Dense Forest, MTF). Spore abundance was lowest in LLF and highest in REF, with no statistical differences relative to MTF. Spore diversity indices and root colonization rates were not statistically different among the phytophysiognomies. However, principal components analysis revealed that AMF community...

Araujo,Ademir Sergio Ferreira de; Mendes,Lucas Wiliam; Bezerra,Walderly Melgaço; Nunes,Luis Alfredo Pinheiro Leal; Lyra,Maria do Carmo Catanho Pereira de; Figueiredo,Marcia do Vale Barreto; Melo,Vania Maria Maciel.
Abstract We used 16S rRNA sequencing to assess the archaeal communities across a gradient of Cerrado. The archaeal communities differed across the gradient. Crenarcheota was the most abundant phyla, with Nitrosphaerales and NRPJ as the predominant classes. Euryachaeota was also found across the Cerrado gradient, including the classes Metanocellales and Methanomassiliicoccaceae.

Biological nitrogen fixation and agronomic features of soybean (Glycine max (L.)Merr.) crop under different doses of inoculant Acta Agron. (Palmira)
Pedrozo,Alessandro; Girelli de Oliveira,Nelson João; Alberton,Odair.
Abstract Soybean is one of the most important crops in the world and presents great versatility. This study aims to investigate the effect of the Bradyrhizobium japonicum inoculant at 0, 600 and 1200 mL ha1 on soybean crop, with and without 1 L ha-1 calcium (Ca 41%) + 0.4 L ha-1 boron (B 14%). Plants were grown for 90 days in plastic pots with 3 kg of sand and vermiculite (v/v) autoclaved at 121oC for 1 h. The experiment was conducted in a completely randomized design with five replications in a greenhouse. Soybean yield parameters were evaluated, such as, dry weight of shoot, root and total; plant height, nitrogen in the shoot; number and dry weight of nodules; chlorophyll index in the leaves; number of pods per plant (NPPL); number of grains per pod...

Fungal Community Structure as an Indicator of Soil Agricultural Management Effects in the Cerrado Rev. Bras. Ciênc. Solo
Valadares-Pereira,Alana de Almeida; Oliveira,Eláiny Cristina Alves Martins; Navarrete,Acacio Aparecido; Oliveira Junior,Waldesse Piragé de; Tsai,Siu Mui; Peluzio,Joenes Mucci; Morais,Paula Benevides de.
ABSTRACT Forest-to-agriculture conversion and soil management practices for soybean cropping are frequently performed in the Cerrado (Brazilian tropical savanna). However, the effects of these practices on the soil microbial communities are still unknown. We evaluated and compared the fungal community structure in soil from soybean cropland with soil under native Cerrado vegetation at different times of the year in the Tocantins State. Soil samples were collected in two periods after planting (December) and in two periods during the soybean reproductive growth stage (February). Concomitantly, soil samples were collected from an area under native Cerrado vegetation surrounding the agricultural area. The soil DNA was analyzed using a fingerprinting method...

Land-use systems affect Archaeal community structure and functional diversity in western Amazon soils Rev. Bras. Ciênc. Solo
Navarrete,Acácio Aparecido; Taketani,Rodrigo Gouvêa; Mendes,Lucas William; Cannavan,Fabiana de Souza; Moreira,Fatima Maria de Souza; Tsai,Siu Mui.
The study of the ecology of soil microbial communities at relevant spatial scales is primordial in the wide Amazon region due to the current land use changes. In this study, the diversity of the Archaea domain (community structure) and ammonia-oxidizing Archaea (richness and community composition) were investigated using molecular biology-based techniques in different land-use systems in western Amazonia, Brazil. Soil samples were collected in two periods with high precipitation (March 2008 and January 2009) from Inceptisols under primary tropical rainforest, secondary forest (5-20 year old), agricultural systems of indigenous people and cattle pasture. Denaturing gradient gel electrophoresis of polymerase chain reaction-amplified DNA (PCR-DGGE) using the...

Metodologia de aplicação de microrganismos solubilizadores de fósforo em sementes visando melhor aproveitamento deste nutriente pelas plantas. Infoteca-e

Sugarcane trash levels in soil affects the fungi but not bacteria in a short-term field experiment BJM
Rachid,C.T.C.C; Pires,C.A; Leite,D.C.A; Coutinho,H.L.C; Peixoto,R.S; Rosado,A.S; Salton,J; Zanatta,J.A; Mercante,F.M; Angelini,G.A.R; Balieiro,Fabiano de Carvalho.
Abstract The sugarcane in Brazil is passing through a management transition that is leading to the abolition of pre-harvest burning. Without burning, large amounts of sugarcane trash is generated, and there is a discussion regarding the utilization of this biomass in the industry versus keeping it in the field to improve soil quality. To study the effects of the trash removal on soil quality, we established an experimental sugarcane plantation with different levels of trash over the soil (0%, 50% and 100% of the original trash deposition) and analyzed the structure of the bacterial and fungal community as the bioindicators of impacts. The soil DNA was extracted, and the microbial community was screened by denaturing gradient gel electrophoresis in two...
